Action mechanism of nicotine on the isolated rabbit ileum.

Abstract
The authors studied the action of nicotine on the isolated rabbit ileum and obtained the following results: 1) Nicotine in low concentration (10-7?10 6 g/ml) caused contraction of the isolated rabbit ileum. In
high concentration (10-5?10-4 g/ml) it caused a transient inhibition followed by stimulation.
2) The "transient pre-inhibitory effect" of nicotine was enhanced, but its excitatory effect was abolished, after atropine.
3) Both the pre--inhibitory and excitatory effects of nicotine were abolished by hexamethonium.
4) The transient pre-inhibitory effect of nicotine was not blocked by either kind of blocking agent alone, but was blocked by a combination of the two.
5) The transient pre-inhibitory effect of nicotine was blocked by bretylium.
6) In the isolated ileum from reserpinized rabbits, the pre-inhibitory effect of nicotine did not appear.
7) The results support the conclusion that there are two kinds of ganglion cells in the myenteric plexus, one set being excitatory and cholinergic, the other inhibitory and adrenergic, and that nicotine stimulates both of these ganglion and produces the biphasic reaction of initial inhibiton followed by contraction.
